SECTION II. APPLIED BIOTECHNOLOGY IN THE PHARMACEUTICAL INDUSTRY... 437glucose concentration of 0-1g/L before reacting with the reagent. Taking 150 %u00b5L of GOD-POD into the reaction well, then taking an exact amount of 15%u00b5L of the diluted sample into the reaction well, mixing well and let the reaction take place in the dark at 300C for 30 minutes. The reaction of the control sample and blank sample was performed with 15mL of glucose solution 1.0 g/L and 15%u00b5L of distilled water. The absorbance of the reaction mixture was measured at the wavelength of 510nm. The glucose concentration of the hydrolyzed solution was determined:In which: Cglucose: Glucose concentration of hydrolysate (g/L)Ameasurement, Acontrol, Ablank: Absorbance of the measurement sample, control sample and blank sample, respectively.f: dilution factorb. Determination of glucose content in hydrolysate by HPLC The cellulose content in bagasse was analyzed in the form of glucan analyzed by HPLC to determine glucose, xylose, arabinose, galactose. The analyzed sample was diluted to the concentration range of the standard curve and then filtered through a membrane 0.2 %u00b5m before being injected into the column. The sample was prepared, analyzed on the system Agilent 1200, column Aminex HPX-87P with a sample volume of 20 %u00b5L. The mobile phase was purified water (Merck, Germany) with a flow rate of 0.6 mL/min. The analysis was performed at 800C with a RID detector and a retention time of 25 minutes. The hydrolysis efficiency was calculated according to the formula:
