SECTION II. APPLIED BIOTECHNOLOGY IN THE PHARMACEUTICAL INDUSTRY... 333wild-type strain. The culture broth of A. hibisca M4 and A. hibisca P172-2 were obtained from these strains incubated in production media at 28%u00b0C, 200 rpm for 7 days. It is shown that the color of A. hibisca M4 culture broth is more dark-red than the color of A. hibisca P172-2 culture broth.Figure 1. The culture broth of A. hibisca M4 and A. hibisca P172-21: culture broth of A. hibisca M4 after incubating2: culture broth of A. hibisca P172-2The quantity of pradimicin production of mutant and wildtype strains was determined by HPLC. The Figure 2 shows the HPLC chromatography of crudes from A. hibisca M4 and A. hibiscaP172-2. The pradimicin production of A. hibisca M4 is 52.4%u00b10.4 mg/L is 6.1-fold higher than that of A. hibisca P172-2 is 8.6%u00b10.2 mg/L (Table 1). To prove the peak is pradimicin A, the crude of A. hibisca M4 was analyzed by LC-MS. Figure 3 shows that the compound in pradimicin A peak (Figure 2) was established at m/z 841.2673 [M + H]+ (calcd., 841.2662). This confirms that the mutant strain M4 produced pradimicin A with a higher yield than the wild-type strain.
