                                    ENHANCING PRODUCTION OF PRADIMICIN A BY RIBOSOME ENGINEERING(Chung Thanh Nguyen*, Huong Thi Nguyen, Thao Thi Phuong Nguyen, Anh Thi Ngoc Nguyen, Khanh Phuong Do, Thoa Kim Vu)1Abstract: Actinomadura hibisca P157-2 is a Gram-positive actinomycete that produces pradimicin A, a pentangular polyphenol compound with antifungal and antiviral activities. The enhancement of pradimicin production is necessary for producing large amount of this compound. In this study, ribosome engineering was performed to develop 20 mutant strains. Among them, 19 mutant strains exhibited pradimicin yields higher than that of the wild-type strain. The highest pradimicin yield achieved by A. hibisca M4 was 52.4%u00b10.4 mg/L. The rpsL gene of A. hibisca M4 was mutated at codon 43 (K43M).Keywords: Actinomadura hibisca P157-2, pradimicin A, ribosome engineering, enhancing production, natural products. 1.
