CLONING AND EXPRESSION OF DGDP- 4,6-HEXOSE DEHYDRATASE/4-EPIMARASE GENE (NCSS) FROM NEOCARZINOSTAIN BIOSYNTHESIS GENE CLUSTER FROM STREPTOMYCES CARZINOSTATINTa Thi Thu Thuy*, Le Thanh Hai Ha, Do Phuong Khanh, Nguyen Thi Huong, Nguyen Thanh Chung1Abstract: The neocarzinostain antibiotic biosynthetic gene cluster isolated from Streptomyces carzinostain was completed. The ncsS is located in the gene cluster and encodes for a peptide of 332 amino acids putatively functioning dGDP- 4,6-hexosedehydratase/4-epimarase that involves in the neocarzinostain biosynthetic pathway. The enzyme shows high homology with UDP-glucose-4-epimerase and TDP-glucose-4,6-dehydratase from Streptomyces strains. For the elucidation of NcsS2 function, the ncsS2 was cloned in pET-32a(+) vector creating pncsS53 as expression plasmid. We have succeded heterologous expressing this gene in E.coli host with His6-tag fusion protein at N- terminal.
